Annotation -
The laboratories are conducted in a 14-day block during March. The exact dates will be announced in the 1st week of the LS. The transfer of theoretical knowledge of the field of ecotoxicology into practice. Students will learn the foundation of selected ecotoxicological tests on organisms of different trophic levels (decomposers, producers, consumers) terrestrial and aquatic environments, methods of evaluation and statistical analysis of the results and their subsequent interpretation.

Students have to graduate all laboratory works and consign protocols from these works and final protocol (comparison of results from all toxicity tests).

Requirements to the exam -

Attendance is mandatory. After exercise students will work out protocols of individual laboratory works within the specified time.

Classification is based on the way the class works, the quality of the reports and their timely submission.

Syllabus -

1. Treatment of samples for toxicity tests.

2. Working with a microscope.

3. Working under sterile conditions.

4. Breeding and cultivation conditions of the test organisms (Daphnia magna, Folsomia candida).

5. Planning of experiments, designing concentration series,

6. Measurement by digital image analysis.

7. Test on brine shrimp (Artemia salina).

8. Growth inhibition test on chlorococcal algae (Desmodesmus subspicatus).

9. Growth inhibition test of aquatic macrophytes - duckweed (Lemna minor).

10.Test of germination and root growth inhibition of mustard (Sinapis alba).

11.Test of inhibition of dehydrogenase activity of soils.

12.Bioluminiscence inhibition test using Aliivibrio fischeri.

13.statistical evaluation of measured data (NOEC, LOEC, EC50).

14.Protocols processing and comparison of results.

  • do basic techniques of manipulation with organisms and obtain basic skill of microscopy
  • perform basic ecotoxicological tests with aquatic and terrestrial organisms of all trophic levels
  • measure basic traits via image analysis
  • perform statistical evaluation and interpretation of ecotoxicological data
  • evaluate protocols on ecotoxicological issues.
