Annotation -
Laboratory exercise from mikrobiology introduces students to elementary work techniques of microbiology. They are instructed in the principal izolation techniques of microorganisms, staining of microorganisms, including Gram staining, and microscopy of yeasts and micromycets. They will learn to evaluate the impact of physical and chemical effects on microbial growth.

Syllabus -

1. Safety rules in microbiological laboratory. Sterilization techniques. Preparation of sterile medium in cultivation flaks and test tubes.

2. Sterilization of pipettes test-tubes and other instruments Defined and komplex media. Inoculation techniques of bacteria and yeasts.

3. Optical microscopy of native and stained yeasts and bacteria. Gram staining of chosen bacterial representatives. Microscopy of different genuses of yeasts, types of their vegetative reproduction.

4. Isolation techniques of microoorganisms, streaking technice for isolation of two bacterial strains from the mixture. Vitality test of yeasts by methylen blue staining.

5. Use of Thoma chamber for enumeration of yeasts. Isolation of pure cultures from the mixture of two yeasts species.

6. Factors influencing bacterial growth:tempetrature, presence of organic dyes, UV irradiation, antibiotics. Continuation of the bacterial and yeasts izolation.

7. Measurement of the growth curve of bacteria (Escherichia coli), calculation of of the specific growth rate by measurement of culture optical density. Time of lag, generation time.

8. Microscopy of micromycets, preparation of native preparate. Comparison of typice representatives of molds(micromycets).

9. Detection of coliform bacteria in food samples. Isolation of bacteria - confirmation of species identity by chosen biochemici tests and by ENTERO-test.

10. Isolation of yeasts species: confirmation of identity by fermentative test and by morfology.

11. Summarization of the results of bacterial izolation. Confirmation of the presence of two bacterial strains.

12. Summarization of the results of yeasts izolation, evaluation of izolation methods.

13. Microscopical identification of anonymous micromycets strain. Confirmation of the presence of two yeasts strains.

14. Disposal of all used microbial cultures, closing test.

Learning outcomes -

Students will be able to prepare microscopic preparates, including diagnostic staining, to determine on the morfology basis the main genuses of yeasts and micromycets. They will know how to interpret ENTEROtest and on the basis of thein own measurement they will be able to calculate the specific growth rate of model microorganisms. They will master the priciples of aseptic work and to prepare basic types of the growth media.
