Annotation -
The course is directed to the knowledge dealing with relations between microorganisms and their environment. Lectures will include information on characterisation of microorganisms, degradation of substrates. Special attention will be paid to the influence of environmental conditions on metabolical and physiological activity, microorganism detection and enumeration. Requirements on microbiological laboratory and general instructions for safe microbiological handling will be mentioned in the last part. Part of the subject will be directed to the laboratory experiments. Students will be introduced into application of basic enumeration techniques for environmental samples.

Aim of the course -

Students will be able to practically apply the knowledge from the field of:

  • microorganism taxonomy,
  • metabolism and genetic,
  • both theoretical and practical basic laboratory techniques used for determination and characterization of microorganisms.

Syllabus -

1. Specification of the subject Microbiology in environment, basic microbiological terms, microbiological taxonomy.

2. Basic structure of microbial cells and their chemical content.

3. Metabolical processes in microorganism.

4. Influence of internal and external factors on growth and characteristics of microorganisms.

5. Nutrition of microorganisms. Growth curve.

6. Principles of both detection and enumeration of microorganisms by cultivation techniques. Sampling and sample storage.

7. Occurence and significance of microorganisms in environment and their detection.

8. Requirements on microbiological laboratory. General instructions for microbiological handling.

9. Laboratory experiments. Safe handling in microbiological laboratory. Preparation of sterile materials.

10. Laboratory experiments. Control of hygiene and laboratory sanitation.

11. Laboratory experiments. Enumeration of bacteria, yeasts and moulds in soil.

12. Laboratory experiments. Enumeration of both culturable and indicator microorganisms in water sample.

13. Laboratory experiments. Isolation techniques. Sample preparation for light microscopy.

14. Laboratory experiments. Testing the effect of external factors on the growth of microorganisms.
