Annotation -
The aim of this course is introduction and mainly practicing of the disconnection approach (= retrosynthetic analysis) in organic synthesis. During the course, the retrosynthetic method will be used for the synthesis of complex structures including total synthesis of natural compounds.

Students will be able to:

Use nomenclature of retrosynthetic analysis

Apply retrosynthetic analysis to design synthesis of polyfunctional organic compounds

Recognize suitable structural elements suitable for disconnection of complex molecules

Understand logic of total synthesis of natural compounds

2. One and two group C-X disconnections.

3. One group C-C disconnections

4. Alkene and alkyne synthesis. Synthesis of biaryls.

5. Synthesis of 1,3- and 1,5-difunctionalised compounds.

6. Synthesis of 1,2-, 1,4- and 1,6-difunctionalised compounds.

7. Strategy of ring synthesis.

8. Synthesis of heterocyclic and heteroaromatic compounds.

9. Chemoselectivity, regioselectivity and stereoselectivity in organic synthesis.

10. Retron, key structural element for complex compound disconnection.

11. Modern methods for transformations of functional groups.

12. Linear and convergent synthesis.

13. Modern approach to total synthesis of natural compounds - recent examples (workshop).

Course completion requirements -

It is compulsory to prepare project on the synthesis of natural compound, to pass written examination test with the result at least 60%, and finally to pass successfuly the oral examination.
