Annotation -
The course covers the basics of chiroptical methods, up-to date instrumentation and methodology of circular dichroism measurements. Variants of chiroptical methods are described together with their advantages, disadvantages and limits. The course is focused on electronic and vibrational circular dichroism and Raman optical activity and their application in structural analysis of chiral molecular systems, in biologically important interactions and chiral recognition.

Aim of the course -

Students obtain:

• theoretical basics of chiroptical spectroscopy

• knowledge of instrumentation of the variants of chiroptical methods

• knowledge of the measurement methodology

• application of chiroptical method in conformational analysis of small molecules and biomolecules

• application of chiroptical spectroscopies in biologically important interactions and chiral supramolecular systems

• participation in the demo measurements of electronic and vibrational dichroic spectra

Syllabus -

1. Molecular chirality, polarization of radiation utilized in chiroptical methods

2. Theoretical basics of chiroptical methods, observable quantities (circular dichroism, ellipticity, optical rotatory dispersion, dissymmetry factor)

3. Interaction of circularly polarized radiation with matter, rotational strength of transition, relation between observables and structure of chiral molecules

4. Methodology of circular dichroism measurements, photoelastic modulator and dedicated devices

5. Electronic circular dichroism, instrumentation, how to obtain reliable spectra

6. Vibrational circular dichroism, its particularity, instrumentation, how to obtain reliable spectra and structural data

7. Variants of chiroptical methods 1: optical rotation and optical rotatory dispersion, fluorescence detected circular dichroism and their instrumentation

8. Variants of chiroptical methods 2: varianty chiroptických metod 2: circularly polarized luminescence, magnetic optical activity and their instrumentation

9. Raman optical activity and its instrumentation

10. Processing chiroptical data, ab initio calculations of chiroptical properties

11. Application of circular dichroism generally and in conformational study of biomolecules

12. Application of circular dichroism in study of biologically important interactions and of chiral supramolecular systems

13. Demo experiments in vibrational circular dichroism laboratory

14. Demo experiments in electronic circular dichroism laboratory
