Annotation -
The transfer of theoretical knowledge of the field of ecotoxicology into practice. Students will learn the foundation of selected ecotoxicological tests on organisms of different trophic levels (decomposers, producers, consumers) terrestrial and aquatic environments, methods of evaluation and statistical analysis of the results and their subsequent interpretation.

Aim of the course -

Students will be able to:

  • do basic techniques of preparation organisms and obtain basic skill of microscopy
  • perform basic ecotoxicological tests with aquatic and terrestrial organisms (microorganisms, aquatic crustaceans, aquatic macrophytes, algae, soil invertebrates, terrestrial plants)
  • perform statistical evaluation and interpretation of ecotoxicological data
  • evaluate protocols on ecotoxicological issues.

Requirements to the exam -

The labs are designed as a 14-day batch. The exact date will be announced before the beginning of the semester.

Attendance is mandatory. After exercise students will work out protocols of individual laboratory works within the specified time.

Syllabus -

1. Preparation and pre-treatment of samples for toxicity tests.

2. Working with a microscope

3. Working in a sterile box.

4. Breeding and cultivation conditions of the test organisms.

5. Planning of experiments, the choice of the concentration series, statistical evaluation of data.

6. Test of acute toxicity to aquatic crustaceans (Daphnia magna).

7. Test on brine shrimp (Artemia salina).

8. Growth inhibition test on chlorococcal algae (Desmodesmus subspicatus).

9. Growth inhibition test of aquatic macrophytes - duckweed (Lemna minor).

10.Terrestrial test - inhibition of root growth of lettuce (Lactuca sativa).

11.Test of inhibition of dehydrogenase activity of soils.

12.Behavioral test with soil invertebrates.

13.Bioluminiscence inhibition test on Vibrio fischeri.

14.Protocols processing and comparison of results
