Annotation -
The aim of the subject is to acquire both theoretical and experimental knowledge of modern laboratory techniques. Among the performed experiments are cross-coupling reactions, photochemical reactions and other reactions that require handling of organometallic and hydride reagents, working under inert atmosphere using inert lines and septum techniques, working in a glovebox, working at low temperatures and with low-boiling liquids. The products obtained will be isolated by advanced separation techniques (vacuum distillation, crystallization and column chromatography) and their structure and purity will be determined by selected analytical methods (NMR spectroscopy, TLC, UV/VIS spectroscopy). Significant part of the course is devoted to the work on research project in the laboratory of student's tutor.

Teaching methods -

Students will attend a laboratory technique course (1st - 4th week of semester, entry 1-8 according to syllabus). Further work will be carried out in specialized laboratories (5th – 14th week of semester). Organization of specialized laboratories will be settled with the student's supervisor.

Syllabus -

1. Determination of unknown concentration of butyllithium solution – working under inert atmosphere using inert lines and septum techniques.

2. Birch reduction of o-xylene – working with low-boiling liquids, working at low temperature, determination of reaction conversion by NMR spectroscopy.

3. Reduction of a carboxylic acid ester – working with hydride reagents under inert atmosphere, crystallization, determination of product purity by NMR spectroscopy.

4. Acylation of cyclohexanone - utilization of enamines in organic synthesis, working with Dean-Stark apparatus, vacuum distillation.

5. Horner-Wadsworth-Emmons reaction – working with sodium hydride under inert atmosphere, crystallization, determination of product purity by NMR spectroscopy.

6. Negishi cross-coupling – working with organozinc reagents under inert atmosphere, column chromatography.

7. Photochemical oxidation of sulfides to sulfoxides – measurement of UV spectra, calculation of extinction coefficient, weighing on an analytical balance, analysis of the reaction mixture by HPLC.

8. Ring-closing metathesis – working in a glovebox, kinetic study using NMR spectroscopy.

9. Research project in a specialized laboratory.
