Annotation -
The course focuses on understanding the whole process of industrial bioprocesses e.g. cell cultivation, upstream and downstream processes, process interactions and engineering aspects of the process. The subject is based on expansion, synthesis and application of knowledge gained in basic subjects such as microbiology, biochemistry, physical chemistry and chemical engineering to bioengineering aspects of industrial bioprocesses. The subject brings new or significantly expanding existing knowledge in the field of mass balance bioprocesses, types of bioreactors and methods of management and control of bioprocesses.

Aim of the course -

Students will be able to:

Apply knowledge of basic subjects such as microbiology, biochemistry and chemical engineering to bioengineering area.

Propose a suitable type of cultivation and bioreactor for the desired product.

Design and implement a mass balance and basic process control.

Syllabus -

1. Structure of the biotechnological process in terms of unit operations. Engineering aspects of the use of intact cells and enzymes as producers.

2. Equipment and unit upstream operations – sterilization of liquid medium and bioreactor, SIP station, filtration and sterilization of gases. Parameters to describe the cultivation.

3. Batch cultivation – characteristics, mass balance, applications.

4. Fed-batch cultivation – characteristics, feed strategies, mass balance, applications.

5. Continuous cultivation – characteristics, process control methods, applications.

6. Continuous cultivation – mass balance, comparison with batch and fed-batch cultivation, process variations.

7. Aeration and oxygen transfer – implementation of aeration, mechanism of oxygen transfer mass balance.

8. Mixing – types of mixing in biotechnology plants, specifics of bioreactor mixing.

9. Bioreactors – classification, design, application; pneumatically and mechanically stirred aerated bioreactors.

10. Bioreactors – photobioreactors, membrane bioreactors, tissue culture reactors, reactors for biotechnology in waste management and environment.

11. Measured and controlled variables in cell cultivation. Basics of bioprocess control – control circuit, types of controllers, variables in control.

12. Equipment and unit downstream operations – cleaning, CIP station, conventional separation technology, cell disintegration.

13. Equipment and unit downstream operations – membrane separation technologies, extraction, precipitation, distillation, drying, stripping.

14. Methods of optimization and scale-up of bioprocesses.
