Annotation -
The aim of the subject is to introduce the molecular biology techniques of forensic analysis of common biological samples. It deals with isolation of nucleic acids, evaluation of its amount and integrity. The key part is the introduction to the techniques of forensic human identification with overlap to laboratory practice.

Aim of the course -

Students will be able to:

understand the basics of DNA and RNA work

isolate nucleic acids from common biological sources

understand the basics of nucleic acid analysis by restriction cleavage

understand the basics of amplification using PCR

understand the basics of quantification of DNA using real-time PCR and digital PCR

understand the basics of human identification using RFPL, VNTR, STR, SNP

1. Introduction, safety in molecular biology laboratory

2. Basics of work with biological material, namely DNA and RNA

3. Isolation of DNA from buccal swab

4. DNA integrity assessment by electrophoresis, measurement of concentration using spectrophotometry

5. Polymerase chain reaction (PCR)

6. VNTR analysis, lenght of the fragments, determination of number of repetitions

7. mt DNA analysis - advantages and restrictions

8. RFLP analysis - cleavage of DNA by restriction endonucleases

9. Electrophoretic separation and analysis of rectriction fragments

10. Nucleic acid quantification using real-time PCR

11. Fundamentals of optimalization of polymerase reactions and primer selection, factors affecting specifity and yield

12. Human identification based on short tandem repetition (STR) analysis I - multiplex PCR

13. Human identification based on short tandem repetition (STR) analysis II - capillary analysis of DNA amplicons
