Annotation -
The course focuses on understanding the whole process of industrial bioprocesses e.g. cell cultivation, upstream and downstream processes, process interactions and engineering aspects of the process. The subject is based on expansion, synthesis and application of knowledge gained in basic subjects such as microbiology, biochemistry, physical chemistry and chemical engineering to bioengineering aspects of industrial bioprocesses. The subject brings new or significantly expanding existing knowledge in the field of mass balance bioprocesses, types of bioreactors and methods of management and control of bioprocesses.

Aim of the course -

Students will be able to:

Apply knowledge of basic subjects such as microbiology, biochemistry and chemical engineering to bioengineering area.

Propose a suitable type of cultivation and bioreactor for the desired product.

Design and implement a mass balance and basic process control.

Syllabus -

1. Biotechnology and bioengineering, biotechnological process structure.

2. Technological aspects of use of intact cells as a producers (plant vs. animal vs. microorganisms). Types of products. Application of enzymes.

3. Upstream processes (storage of microorganisms, preparation of inoculum, preparation and sterilization of liquid media, sterilization of reactor, SIP, filtration and sterilization of gases).

4. Parameters for the description of microorganism cultivation. Yield and productivity of process. Overview of cultivation types.

5. Batch cultivation (characteristics, mass balance, use).

6. Fed-batch cultivation (characteristics, types of feeding, mass balance, use).

7. Continuous cultivation (characteristics, process control, mass balance, compared to batch cultivation, use). Choosing the cultivation type on the basis of the desired product.

8. Aeration and oxygen transfer (implementation of aeration, oxygen transfer mechanism, KLa).

9. Mixing (mixing types in biotechnological plant, specifics of mixing in bioreactors).

10. Bioreactors (types, design, use).

11. Measured and controlled variables in cell cultivation. Fundamentals of bioprocess control (controllers, control loop, variables in controlling).

12. Downstream processes (conventional separation technologies, disintegration of cells, membrane separation technologies).

13. Downstream processes (extraction, precipitation, distillation, drying, stripping, CIP). Production line of biotechnological process.

14. Biotechnologies and bioreactors for treatment of wastes from biotechnological processes.
