Amino acids (AA), their codes and basic properties. AA derivatives, orthogonally protected and their utilization in peptide synthesis in solution. Coupling agents and some side reaction, occurring during condensation. Some other synthetic difficulties in peptide synthesis. Examples of peptide preparation. Peptides used as therapeutics and their production. Survey of the chosen peptides - active pharmaceutical ingredients (API). Basic principles of solid phase synthesis. Paralell synthesis, robotics, and construction of the libraries. Utilization of combinatorial chemistry and future of peptides.

describe methods of preparation of synthetic peptides by various classical and modern procedures

Describe basic methods of their analysis and technology of production

View peptides as a group of natural compounds with a chance of their structural modifications with respect to biological and pharmacological properties and further utilization in medicine

1. Amino acids (AA)

2. Protecting groups: protection of amino and α-carboxylic groups. Tactics for protection of amino acids side chains.

3. Coupling alternatives in carboxylic group activation, frequently used coupling agents, methods of condensation of AA derivatives, interesting and frequent side reactions.

4. Other methods of amide bond formation, chemical ligation and use of enzymes for peptide synthesis.

5. Examples of peptide synthesis in solution, analysis, and commercial peptide production.

6. Peptide drugs: biological properties of chosen peptide, production methods - their limits and advantages, survey of the main peptide pharmaceuticals.

7. Solid phase peptide synthesis concept.

8. Automatic peptide synthesis and design of the synthesizers.

9. Parallel peptide synthesis.
