The aim of the subject is to introduce the molecular biology techniques of forensic analysis of common biological samples. It deals with isolation of nucleic acids, evaluation of its amount and integrity. The key part is the introduction to the techniques of forensic human identification with overlap to laboratory practice.

understand the basics of DNA and RNA work

isolate DNA from common sources

understand the basics of nucleic acid analysis by restriction cleavage (RFLP)

understand the basics of quantification of DNA using real-time PCR

understand the basics of human identification using VNTR and STR polymorphism

understand the basics of DNA sequencing and fragment analysis

1. Introduction, safety in molecular biology laboratory

2. Basics of work with biological material, namely DNA

3. Isolation of DNA from buccal swab, quality and quantity assessment

4. Polymerase chain reaction (PCR) - amplification of tandem repeats (locus D1S80)

5. Electrophoretic separation of amplicons

6. Determination of number of repeats (locus D1S80) and random match probability (RMP)

7. Amplification of mitochondrial DNA (mtDNA) using PCR

8. RFLP analysis of mtDNA - cleavage of DNA by restriction endonucleases
