The course provides introduction to the world of macromolecules, covers brief overview of polymer physics and chemistry with special focus on contemporary polymer research and technology in biomedical field.
Aim of the course -

Students will gain the following skills:

1. understand the fundamental physical concepts governing the behavior of polymers in solutions

2. design chemical synthesis of various synthetic polymers including control of their chain lengths

3. recognize parameters governing the interaction between a polymer and living system on both cellular and whole body level

4. follow recent trends in biomedical applications of polymers in emerging field such as tissue engineering, drug delivery and 3D cell cultures

1. Polymery – úvod: definice, fyzikální a chemické vlastnosti, syntetické-přírodní polymery, aplikace, životní cyklus

2. Fyzika polymerů I – Fraktální dimenze, ideální a reálné řetězce, termodynamika polymerů v roztocích, Flory Huggins teorie

3. Fyzika polymerů II – Charakterizace polymerů: optická, chromatografická, osmometrie, elektrokinetika

4. Chemie polymerů – Polykondenzace, polymerace, kopolymery, kopolymerní faktory, emulzním suspenzní polymerace, ATRP, RAFT

5. Polymery reakční inženýrství I – úvod do metody momentů, kinetika volné radikálové polymerace, Trommsdorff-Norrish efekt, “ceiling temperature”

6. Polymery reakční inženýrství II – kinetika polykondenzace, gelace

7. Polymerní chemie II – kopolymery, „star“ polymery, dendrimery, gely – fyzika (perkolace, gelační bod, rovnovážné botnání), typy síťování, viskoelasticita, rheologie

8. Syntetické polymery pro biomedicínské aplikace – akryláty, PEG, PCL, PLA, implantáty, excipienty, degradace

9. Biopolymery – typy (polysacharidy, proteiny, nukleové kyseliny), funkce v živých systémech, metabolismus, zdroje, chemická modifikace, aplikace

10. Interakce organizmus-polymer na buněčné úrovni: povrch buňky, internalizace, transfekce, buněčný metabolizmus

11. Interakce organizmus-polymer na úrovni celého organizmu: farmakokinetika, resorpce z krve, EPR efekt, transport tkání, akumulace, exkrece, interakce s imunitním systémem

12. Polymery v drug delivery – farmakokinetika vs. velikost, možnosti uvolnění léčiva

13. Polymery v medicíně – ortopedie, oftalmologie, zubařství, onkologie, nefrologie, ochrana zranění

14. 3D buněčné kultury, regenerativní medicína, tkáňové inženýrství
