Annotation -
The course is focussed on a deeper understanding of analytical liquid chromatgraphy and gas chromatography, with the emphasis on combination with mass spectrometer as a detector. Students will be acquainted with the theoretical principles of liquid and gas chromatography, optimization of both methods, practical examples and interpretation of mass spectra.

Aim of the course -

Students will understand both the theoretical basis and the practical aspects of liquid and gas chromatography combined with mass spectrometry. They will be able to solve and develop tasks related to the optimization of gas and liquid chromatographic separation and their subsequent detection and interpretation of mass spectra.

Syllabus -

The course deals with the theoretical principles of chromatography, such as thermodynamic and kinetic theory, elution characteristics, integration, mechanisms of separation. These theoretical foundations are followed by instrumentation in gas and liquid chromatography. The attention is paid to the respective types of detectors. The practical part deals with: a) GC - modification of carrier gas, overview of stationary phases, characteristics of capillary columns and principles of their treatment, temperature program, derivatization; b) LC - selection of stationary and mobile phases, optimizing parameters and optimization of the methods, gradient chromatography.

The other part of the course deals with mass detection: theory, instrumentation, types of ion sources and analyzers, ionization methods, combination of LC/MS and GC/MS, interpretation of the mass spectra. Finally, the course deals with the problematics of the experiment run and also practical examples
