Annotation -
The students will learn modern laboratory techniques in both theoretical and practical way. The course will concentrate on advanced glass-blowing techniques, work under inert atmosphere using simple inert lines and septum techniques, work at low temperature, work under pressure in glass ampules, hydrogenation and work with low-boiling liquids and gases. The products obtained will be isolated by advanced separation techniques (microdistillation, column chromatography) and their structure and purity will be determined by selected analytical methods (NMR spectroscopy, TLC, UV/VIS spectroscopy). Significant part of the course is devoted to the work on research project in the laboratory of student's tutor.

Teaching methods - Czech

V rámci tohoto předmětu posluchači absolvují týdenní kurz laboratorní techniky (organizováno turnusově Po-Pá 8:00 � 17:00, celkem 45 hodin). Další náplní kurzu je práce ve specializovaných laboratořích a literární rešerše (8 hodin týdně � 12 týdnů). Organizaci specializovaných laboratoří si studenti dohodnou s vedoucím práce.

Syllabus -

1. Advanced glass-blowing techniques, making Schlenk flask.

2. Protecting of propargyl alcohol with dihydropyran, control of purity of the product by NMR spectroscopy.

3. Alkylation of protected propargyl alcohol with unknown alkyl iodide,identification of the product by NMR spectroscopy.

4. Deprotection of protected alkynyl alcohol, control of purity of the product by NMR spectroscopy.

5. Determination of unknown concentration of butyllithium.

6. Work under low temperature, synthesis of unknown kinetic silyl enol ether, isolation of the product by microdistillation, identification of the product by NMR spectroscooy.

7. Work in sealed ampules, synthesis of unknown thermodynamic silyl enol ether, isolation of the product by microdistillation, identification of the product by NMR spectroscooy.

8. Katalytic hydrogenation of alkynol to cis-alkenol.

9. Work with low-boiling liquds, reduction of alkynol with sodium in liquid ammonia.

10. Work on the research project in the tutor laboratory.
